Indication:

5HTP is indicated for the treatment of-

• Primary Insomnia and Sleep Architecture Disturbances (Delayed sleep onset, frequent nocturnal awakenings)
• Generalized Anxiety Disorder (GAD), Chronic Stress, and Nervous Tension
• Mild to Moderate Depression and Seasonal Affective Disorder (SAD)
• Fibromyalgia Symptom Relief (Chronic widespread muscle pain associated with low serotonin)
• Chronic Tension Headaches and Migraine Prophylaxis

Dogase & Adminstration:

Adult Dose:

• For Sleep Disorders/Insomnia: Take 1 to 2 capsules orally once daily, 30 to 45 minutes before bedtime, or as directed by a registered healthcare professional.
• For Anxiety and Daytime Stress Relief: Take 1 capsule daily with a glass of water, preferably with a light meal.

Pediatric Dose (Children):

• Children under 12 years: Strictly contraindicated unless explicitly prescribed by a medical specialist.
• Adolescents (12 to 17 years): Safety and efficacy have not been fully established for OTC use; consult a pediatrician for personalized dosing protocols.

Pharmacology:

The pharmacological efficacy of this preparation relies on the dual-pathway mechanism targeting the major inhibitory and regulatory neurotransmitter systems within the Central Nervous System (CNS).

• 5-Hydroxytryptophan (5-HTP) Pathway: Oral 5-HTP readily crosses the blood-brain barrier (BBB) without requiring competitive transport carriers. Once inside the brain, it undergoes rapid decarboxylation via the enzyme aromatic L-amino acid decarboxylase to directly yield Serotonin (5-hydroxytryptamine / 5-HT). By replenishing and stabilizing serotonin pools, it significantly enhances mood, decreases panic responses, and serves as the primary direct biological precursor to Melatonin, which triggers normal sleep onset and maintains biological circadian rhythm.
• Valerian Extract Pathway: Valerian contains bio-active constituents including valerenic acid, valepotriates, and essential oils. It directly targets the GABAergic system. It exerts a triple action: it stimulates the pre-synaptic release of Gamma-Aminobutyric Acid (GABA), blocks its reuptake back into the synaptic cleft, and inhibits the enzymes (such as GABA transaminase) responsible for breaking down GABA. The resulting elevated concentration of GABA increases chloride ion influx, hyperpolarizing neurons and exerting a profound, non-addictive sedative, anxiolytic, and anti-spasmodic effect.

Side Effects:

This natural formulation is generally well-tolerated. However, some individuals may experience:

• Gastrointestinal mild distress (Nausea, vomiting, heartburn, stomach cramps, or diarrhea)
• Daytime drowsiness, dizziness, or a mild hangover feeling (if taken too late at night)
• Vivid dreams or mild headaches.

Drug Interaction:

• Antidepressants (SSRIs, SNRIs, Tricyclics, MAOIs): Concurrent use with Fluoxetine, Sertraline, Amitriptyline, or Phenelzine may lead to dangerous accumulation of serotonin, causing Serotonin Syndrome (marked by hyperthermia, tachycardia, and rigidity).
• Carbidopa: Combined administration may significantly elevate peripheral 5-HTP levels, increasing the risk of developing scleroderma-like skin conditions.
• CNS Depressants & Sedatives: May heavily potentiate the sedative and hypnotic effects of Barbiturates, Benzodiazepines (e.g., Diazepam), and Alcohol.
• Anesthetics: May interact with surgical anesthesia; discontinue use at least 2 weeks before scheduled surgeries.

Pregnancy & Lactation:

• Pregnancy: Strictly not recommended. There is insufficient clinical data to confirm the safety of 5-HTP and Valerian on fetal development.
• Lactation (Breastfeeding): Avoid use during breastfeeding as it is unknown whether the active metabolites pass into breast milk or affect the nursing infant.

Over Dosage:

• Symptoms: Extreme drowsiness, severe nausea, vomiting, dizziness, confusion, and signs of mild serotonin overstimulation (sweating, rapid pulse).
• Management: Discontinue the supplement immediately. Symptomatic and supportive treatment should be initiated. Seek immediate medical attention or stomach lavage if a massive quantity is ingested.
